L'oxydation spontanée des corps gras au contact de l'air donne naissance, lorsqu'elle est notable, à de très nombreux dérivés oxygénés des acides gras, en particulier à des aldéhydes. Ceux-ci réagissent avec la p-anisidine pour donner des composés colorés. L'intensité de la couleur obtenue est proportionnelle à la teneur en aldéhydes et permet donc d'évaluer l'état d'oxydation du corps gras.
